Tato část ISO 12228 specifikuje postup pro stanovení obsahu a složení sterolů v živočišných a rostlinných tucích a olejích plynovou chromatografií. Nicméně stanovení obsahu a složení sterolů v olivovém oleji a oleji z olivových výlisků se provádí s použitím ISO 12228-2. Zkušební podíl se saponifikuje varem pod zpětným chladičem v ethanolickém roztoku hydroxidu draselného. Nesaponifikovatelná hmota se izoluje extrakcí na pevné fázi na koloně s oxidem hlinitým. Kolona s oxidem hlinitým se použije k zachycení aniontů mastných kyselin; steroly projdou kolonou. Sterolový podíl z nesaponifikovatelné hmoty se oddělí pomocí tenkovrstvé chromatografie. Kvalitativní a kvantitativní složení sterolových frakcí se stanoví plynovou chromatografií pomocí cholestanolu nebo betulinu jako vnitřního standardu
