Tato část ISO 12966 specifikuje metodu rychlé transesterifikace tuků a olejů pomocí bazické katalýzy s použitím hydroxidu trimethylsulfonia (TMSH), pro přípravu methylesterů mastných kyselin. Tato metoda je použitelná výhradně pro přípravu methylesterů tuků a olejů pomocí plynové chromatografie s kapalnou stacionární fází (GLC). Je použitelná pro všechny tuky a oleje, s výjimkou těch, které pocházejí z mléka a mléčných výrobků. K izomerizaci nenasycených mastných kyselin dochází pouze v malém rozsahu a izomerované mastné kyseliny jsou pak přítomny jen na úrovni meze stanovitelnosti. Protože dochází k izomerizaci, nedoporučuje se tato metoda pro konjugovanou kyselinu linolovou (CLA)
