Norma uvádí formální a obsahové požadavky na referenční postupy měření. Ty jsou nutné pro získání použitelných a spolehlivých výsledků měření při službách rutinních zdravotnických laboratoří, ale i ve vědě, výzkumu a technice. Prostřednictví referenčních postupů měření se zajišťuje srovnatelnost výsledků a jejich návaznost na jednotky meření, etalony a postupy měření vyšší metrologické úrovně. Obsah normy je rozdělen do 4 kapitol a obsahuje 2 informativní přílohy, z nichž první se zabývá referenčními postupy pro jiné než diferenciální a racionální veličiny a druhá uvádí vzájemný vztah mezi touto normou a Směrnicí EU 98/79/EC. Norma nahrazuje ČSN EN 12286 z února 2000.
