Dieses Dokument legt ein Real-time PCR-Verfahren für die Quantifizierung der Menge an Equiden-DNA im Verhältnis zur gesamten Säugetier-DNA in einer Probe von rohem Fleisch fest. Die Ergebnisse des Equiden-Assays werden in Form von Kopienzahlen des haploiden Genoms von Equiden (Gattung Equus) im Verhältnis zu den Kopienzahlen des haploiden Genoms von Säugetieren insgesamt ausgedrückt. Dieses Verfahren ist spezifisch für Vertreter der Gattung Equus und weist daher DNA von Pferd, Maultier, Esel und Zebra nach. Das Verfahren wurde zuvor in einem Ringversuch validiert und für DNA von Proben aus rohem Pferdefleisch in rohem Rindfleisch als Hintergrund angewendet. Sowohl für die Equiden- als auch für die Säugetier-PCR wurde experimentell eine Nachweisgrenze von mindestens 17 haploiden Genomäquivalenten (HGE; en: haploid genome equivalents) bestimmt. Sie wurde in Einzellaborvalidierung auf Basis der niedrigsten Verdünnung der jeweiligen Kalibrierkurven ermittelt. Der niedrigste im Ringversuch geprüfte relative Pferdeanteil war ein Massenanteil von 0,1 %, basierend auf gravimetrisch vorbereiteten rohem Pferde-Muskelgewebe in rohem Rinder-Muskelgewebe als Hintergrund. Der Prozess der Konformitätsbewertung ist nicht Teil dieses Dokuments.
