Dieser Teil von ISO 21427 legt ein Verfahren zur Bestimmung der Gentoxizität von Wasser und Abwasser mit einem In-vitro-Test mit Säugetierzellen fest. Mit dem Verfahren können durch Wasserinhaltsstoffe verursachte zytogenetische Schäden an den Chromosomen oder am Mitoseapparat der V79-Zellen des Chinesischen Hamsters nachgewiesen werden. Die dabei entstehenden Mikrokerne können Chromosomenbruchstücke und/oder ganze Chromosomen beinhalten. Test basiert auf einer Zunahme der Häufigkeit von mikrokernhaltigen Zellen infolge der Einwirkung von erbgutverändernden Substanzen mit und ohne Zugabe eines metabolisierenden Systems.
